Anwendungen

Endonuclease that splits phosphodiester linkages, preferentially adjacent to a pyrimidine nucleotide and removal of DNA from protein samples. Also used in is used to nick DNA, as a first step to incorporate labeled bases into DNA. The enzyme from Sigma has been used in the nuclease stock along with RNAse during cell lysate preparation of Madin-Darby canine kidney (MDCK) II cell lines.

Bemerkungen

One unit causes an increase in absorbance at 260nm of 0.001 per minute per ml at 25 degrees when acting upon highly polymerized DNA at pH 5.0. Minimum 2,000 units per ml. Solution at 1 mg/ml containing 50% glycerol and 1 mM calcium chloride.
