Applications

Ampicillin is used as asemi-synthetic antibiotic which has antibacterial activity structurally related to penicillin. It is widely used selection reagent for transformed cells expressing β-lactamase. It is also used to prevent and treat a number of bacterial infections. It may also be used to prevent group B streptococcal infection in newborns.

Notes

Stock solution:

An ampicillin stock solution can be prepared at a concentration of 100 mg/mL and should be stored at -20⁰C.

  1. Add 1g of ampicillin to 10mL of dH2O
  2. Sterilize the solution using a 0.22μm filter
  3. Store solution in different aliquots at -20⁰C

LB-ampicillin agar preparation:

  1. Dissolve the following in 500 mL dH2O:
    1. 5g tryptone
    2. 2.5g yeast extract
    3. 5.0g NaCl
    4. 7.5g agar
    5. 25mg ampicillin

    OR

    1. 20g pre-mixed LB agar powder
    2. 25mg ampicillin
  2. Boil solution on stirring hot plate for 1 - 2 min.
  3. Autoclave for 20 minutes and let cool to 50-60⁰C.
  4. Pour approximately 10mL of molten LB agar into each plate.
  5. Allow plates to solidify for approx. 20 min
