The HPLC solvents are highly purified and filtered through 0.2 micron filters. HPLC is classified as normal or reverse-phase based on the type of stationary phase and solvent used to achieve separation. In normal phase HPLC, the stationary phase is polar and the solvents used are non-polar, whereas, in reverse phase the stationary phase is nonpolar and the solvent is polar. HPLC requires both polar and non-polar solvents to achieve separation and analysis. Also, additives are sometimes required in HPLC analyses to improve the peak shape, sensitivity, sample load tolerance, or to enhance chromatographic separation. Common additives are buffers and ion-pairing agents that control the pH of the medium. The most frequently used buffers/additives in proteomics analysis are acids such as formic acid, acetic acid, and trifluoroacetic acid. Basic buffers such as triethylamine and tetrabutylamine are also commonly used. Alfa Aesar supplies highly purified HPLC solvents for liquid chromatography applications.
